Chapter 10.2, Problem 2TQ
Summary Introduction

To review:

The effects of null mutations on the induction of beta-galactosidase by lactose, expression of the lacZ gene, and catabolite repression.

Introduction:

The role of lacI (lactose inducer) is to repress the lac operon. It is a tetramer form that binds to the two-lactose operator region (lacO and lacO1). These operator regions cover the lacZYA promoter and are responsible to control the transcription of these structural genes. LacI on binding prevents the RNA (ribonucleic acid) polymerase activity and stops the transcription process.

What effect does binding of the IRF protein to the IRE in the mRNA encoding ferritin have on the production of ferritin? Briefly explain why this effect is observed.
Explain why in cells that are genetically NF1–/–, basal levels of GTP-bound activated Ras are higher than normal and respond to growth factor stimulation by increasing rapidly to far higher levels.
GC (Glycocorticoids) binds to the glucocorticoid receptor (GR), which leads to the transcription of a series of genes (PC, PCK1, FBP1, PFKFB1, G6PC and G6P transporter (SLC37A4)) involved in gluconeogenesis. What type of receptor is GR? Where would you expect the activated GR to be located to enhance the expression of the gluconeogenic genes? Which cis-acting elements are used by GR as an activator to turn gene transcription “on”?
