Chapter 28, Problem 1CT
Summary Introduction
To review:
The reason that the colonies growing in the close association are smaller than those that are well separated.
Introduction:
Agar is the most commonly used solidifying agent for any culture media. Bacteria can be grown successfully in nutrient-rich agar medium. Bacteria obtain nutrients from the mediumand expand in size. For multiplication and identification of bacteria, this method is usually applied. The given figure below depicts growth of the bacterial colonies on an agar plate. Some of them form into single large cells while others grow into small colonies.

- To observe T. spathacea cells, a sharp blade was used to peel off a thin portion of the lower epidermisof the leaf and was placed on a clean glass slide. One to two drops of 0.01 M NaCl solution was added and a coverslip was placed. The procedure was repeated using 0.30 M NaCl. The leaves were examined under LPO and HPO. A portion with only a single layer of cells was observed. Cells under HPO are shown in your worksheet. Give a short description of the T. spathacea leaf cells in 0.01 M and 0.30 M NaCl solutions. Once a batch of production is done, these cells along with their support material are separated from the medium, and then sterilization is performed in a furnace for safe disposal of material into the environment. An important issue here is to ensure ALL bacteria, including any spores that may form and those that are located at the center of the spherical support material, are killed with heat. For this purpose, the temperature at the center of the spherical support material must reach the designated killing temperature. The support material is a sphere with 3 cm diameter and a heat capacity of 4.0 kJ/kg*K, a density of 800 kg/m³, a thermal conductivity of 0.7 W/m*K. The heat transfer coefficient of the surrounding air is 14 W/m2*K.
